Interstitial lung disease and pneumonitis
Osimertinib can cause ILD/pneumonitis. Beacon’s product information states that confirmed ILD/pneumonitis requires permanent discontinuation.
New or worsening respiratory symptoms should be medically assessed promptly.
QTc prolongation
Osimertinib can prolong the QT interval. Significant QTc abnormalities may require treatment interruption, dose reduction or discontinuation depending on severity.
Cardiomyopathy
Osimertinib can affect cardiac function. Beacon’s prescribing information provides specific management recommendations for reductions in left ventricular ejection fraction and symptomatic heart failure.
Ocular toxicity
Keratitis and other ocular adverse effects can occur. New eye pain, redness, light sensitivity or visual disturbance should be evaluated.
Pregnancy
Osimertinib can cause fetal harm. Pregnancy and reproductive considerations should be discussed with the treating healthcare professional before treatment.
Drug interactions
Strong CYP3A inducers can lower osimertinib concentrations. Beacon identifies medicines such as rifampicin, phenytoin and carbamazepine among examples to avoid.
Other medicines
Osimertinib may also affect concentrations of certain sensitive CYP3A, BCRP or CYP1A2 substrates with narrow therapeutic indices. A complete medication review is therefore important.
